Isolation and characterization of polyhydroxyalkanoate (PHA) producing bacteria isolate from landfill land of Kampung Jawa Banda Aceh

The production of biodegradable plastic from microorganisms has great potential as a substitute for conventional plastic. This study aims to isolate bacterial strains capable polyhydroxyalkanoates (PHAs) the Kampung Jawa landfill land (KJLL) and characterize biopolymers. were able produce PHA using mineral salt medium (MSM) with glucose carbon source. qualitative screening PHA-producing bacteria was conducted by Sudan Black Nile Red. Of 64 strains, only 41 accumulate in results showed that one Coccobacillus strain had highest color intensity further characterization PHA. Differential Scanning Calorimetry (DSC) Thermogravimetric Analysis (TGA) melting temperature (Tm) 101.54°C. X-ray diffraction (XRD) analysis revealed crystalline structure an index crystallinity (ICr) value 15.82% proved produced isolate. suggests this is first report producing good characteristics biotechnology applications.
